DOSAGE

  • Narcolepsy and OSA: 200 mg, it can be taken once a day in the morning.
  • In the case of Shift work sleep Disorder (SWSD) 200 mg can be taken approximately 1 hour before the start of the work shift.
  • Some clinicians may adjust to 100 mg/day or up to 400 mg/day in divided doses based on response and tolerability.

  • Absorption rapidly, peak plasma concentration in just 2-4 hours.
  • Well absorbed. Not significantly affected by food except delayed Tmax.
  • Extensively metabolized in the liver, primarily via CYP3A4/5, and hydrolysis. Main inactive metabolite is modafinil acid. Also involves CYP2C19.
  • Half-life is approx. 12-15 hours for the racemic mixture. The R-enantiomer has longer half-life is approx. 15 hours. This long half-life supports once-daily dosing but can affect night time sleep if taken late.
  • Excretion: Primarily via urine as metabolites
  • Steady State: Reached in 2-4 days of daily dosing.
